
Technology
The DArT genotyping technology consists of several steps:
- Cutting DNA from reference organism(s) and amplification of a set of restriction fragments
- Generation of a library of these restriction fragments
- Printing the library on glass slides, giving micro-arrays
- Cutting DNA from target organisms and amplification of a set of restriction fragments
- Adhering fluorescent labels to these fragments
- Hybridization of the labeled fragments onto the micro-arrays
- Scanning
- Data analysis
Differences in hybridization patterns of target organisms reflect genetic differences between these organisms.
Equipment
Plant Research International has a good infrastructure for performing DArT such as micro-array facilities and colony picker and experienced highly qualified staff. Plant Research International developed a flexible, highly reproducible method for generation of sets of amplified restriction fragments for DArT.
